Formulación de biomezclas para la remoción de plaguicidas de uso agrícola. Evaluación de la bioaumentación con actinobacterias

dc.description.abstractLa atrazina (ATZ) es uno de los herbicidas más utilizados en el mundo y en la región del noroeste argentino, y se ha detectado su presencia en suelos, aguas superficiales y subterráneas. En este contexto, los sistemas de biopurificación (SBP) constituyen una alternativa para prevenir la contaminación por fuentes puntuales asociadas al manejo inadecuado de plaguicidas. Una estrategia para optimizar la eficiencia de un SBP es la bioaumentación con microorganismos con capacidades catabólicas especializadas, entre los que se destacan las actinobacterias. El objetivo general del presente trabajo de Tesis Doctoral fue remover plaguicidas de uso corriente en cultivos regionales mediante biomezclas sustentables y evaluar el efecto de su bioaumentación con actinobacterias. Se evaluó la tolerancia de catorce actinobacterias para crecer y remover ATZ. Todas las cepas demostraron tolerancia al herbicida en concentraciones de 1 g L⁻¹ y fueron capaces de removerlo en medio mínimo con eficiencias de 9,6% a 75,9%. Entre ellas, se seleccionó Streptomyces sp. M7 para la bioaumentación de las biomezclas, por su alta eficiencia de remoción y crecimiento robusto en presencia de ATZ, con niveles de biomasa comparables al control con glucosa. Posteriormente, se formularon tres biomezclas utilizando subproductos lignocelulósicos de la industria azucarera regional (bagazo, cachaza y residuo agrícola de cosecha), combinados con turba y suelo. En ensayos de microcosmos, la bioaumentación con Streptomyces sp. M7 mejoró significativamente la eficiencia de remoción del plaguicida en las tres biomezclas. La cinética de degradación de ATZ se ajustó a un modelo de segundo orden, con disminuciones significativas en la vida media del herbicida en presencia de la actinobacteria. La concentración de inóculo óptima fue de 2 g kg⁻¹, y la inmovilización de Streptomyces sp. M7 en bagazo demostró ventajas en términos de viabilidad celular y eficiencia de remoción. Se seleccionó la biomezcla formulada con bagazo para ensayos a escala de mesocosmos, debido a su comportamiento fisicoquímico y microbiológico favorable, con múltiples aplicaciones de ATZ y sucesivas inoculaciones con Streptomyces sp. M7. En tales ensayos, las biomezclas bioaumentadas mostraron una eficiencia de remoción superior (80–85%) en comparación con las no inoculadas (70–75%). Además, las biomezclas bioaumentadas mantuvieron una mayor diversidad microbiana y una respuesta funcional más estable; sus perfiles fisiológicos comunitarios (CLPP) revelaron una mayor utilización de fuentes de carbono complejas, lo cual sugiere una adaptación funcional al estrés inducido por el herbicida. Se diseñaron SBP con la biomezcla seleccionada y una cobertura vegetal con césped, la cual contribuyó a atenuar la toxicidad de las biomezclas contaminadas,tanto bioaumentadas como sin bioaumentar. Las plántulas de césped crecidas en biomezclas bioaumentadas presentaron mayor vigor y menores niveles de estrés oxidativo. Además, la cobertura vegetal de los SBP contribuyó a atenuar la fitotoxicidad de sus biomezclas sobre plántulas de lechuga, alcanzando valores de elongación radicular e índices de germinación significativamente mayores en los SBP bioaumentados. Los resultados obtenidos demuestran que la combinación de bioaumentación con Streptomyces sp. M7 y la cobertura vegetal es una estrategia efectiva para potenciar la remoción de ATZ en SBP formulados con subproductos agrícolas. es
dc.description.abstractAtrazine (ATZ) is one of the most widely used herbicides worldwide and in the Northwest region of Argentina; its presence has been detected in soils, and in surface and groundwater. In this context, biopurification systems (BPS) represent an alternative to prevent contamination from point sources associated with improper pesticide handling. One strategy to optimize BPS efficiency is the bioaugmentation with microorganisms possessing specialized catabolic capabilities. In this regard, actinobacteria stand out for their potential to biodegrade pesticides, their metabolic diversity, and their ability to colonize various substrates. The general objective of this Doctoral Thesis was to remove commonly used pesticides in regional crops by sustainable biomixtures and to evaluate the effect of the bioaugmentation with actinobacteria. Fourteen actinobacteria were evaluated for their tolerance to grow and degrade ATZ. All strains exhibited tolerance to the herbicide at concentration of 1 g L⁻¹ and were able to remove ATZ in minimal medium with efficiencies ranging from 9.6% to 75.9%. Among them, Streptomyces sp. M7 was selected for bioaugmentation of the biomixtures due to its high removal efficiency, robust growth in the presence of ATZ, and biomass levels comparable to glucose control. Subsequently, three biomixtures were formulated using lignocellulosic by-products from the regional sugar industry (bagasse, sugarcane filter cake, and harvest residue), combined with peat and soil. In microcosm assays, bioaugmentation with Streptomyces sp. M7 significantly improved the pesticide removal efficiency in the three biomixtures. The degradation kinetics of ATZ followed a second-order model, with significant reductions in the half-life of the herbicide in the presence of the actinobacterium. The optimal inoculum concentration was 2 gkg⁻¹, and immobilization of Streptomyces sp. M7 on bagasse demonstrated advantages in terms of cell viability and removal efficiency. The biomixture formulated with bagasse was chosen for mesocosm-scale assays due to its favorable physicochemical and microbiological behavior, with multiple applications of ATZ and successive inoculations of Streptomyces sp. M7. In such assays, the bioaugmented biomixtures showed superior removal efficiency (80–85%) compared to those not inoculated (70–75%). In addition, the bioaugmented biomixtures maintained greater microbial diversity and a more stable functional response; their community physiological profiles (CLPP) revealed higher use of complex carbon sources, which suggests a functional adaptation to the stress induced by the herbicide. Biopurification systems were designed with the selected biomixture and a grass-based vegetative layer, which enabled them to mitigate the toxicity of the contaminated biomixtures, both with and without bioaugmentation. Grass seedlings grown in bioaugmented biomixtures exhibited greater vigor and lower levels of oxidative stress. Additionally, the vegetative layer in the BPS contributed to reducing the phytotoxicity of the biomixtures on lettuce seedlings, achieving significantly higher values of root elongation and germination indices in the bioaugmented BPS. The results obtained demonstrate that the combination of bioaugmentation with Streptomyces sp. M7 and a grass layer is an effective strategy to enhance ATZ removal in BPS formulated with agricultural by-products.en
